When Tony greedily made Ralph give him that giant cut of the winnings he made the horse too low profit to be sustainable, and basically ensured some kind of alternate scheme to make money with the horse. Tony killed that Horse.
I don't think Ralphie killed the horse. Tony accusing Ralphie of killing the horse and not thinking that Ralphie could not have possibly done it just further homes that Tony loves animals more than people and the fact that he's continuing to lose his grip on his crew and reality. As the show goes on he is the one that causes the downfall of his crew. Tony being greedy about the horse causing Ralphie to kill the horse does make sense, but I also think it's funnier that Ralphie is innocent in that particular situation.

What matters here is that Tony knew he was giving Ralph motive to fuck him over when he was an asshole over those horse race winnings, only his response isn't to stop being an asshole, it's to resent Ralph for maybe now having a motive & a specific way to get back at him.
Tony's already so bitter that he can only muster up any empathy for truly innocent creatures, like animals, so he's distraught by the horse he cared about dying and wants to feel like it's the fault of someone he can punish.
Ralph is a slimeball shitheel that Tony still hates for beating that dancer to death, so Tony wants the justification of a horse murder so badly it makes him ignore reality
Then the icing on the cake is Ralph using the same whiny explanation he used for why him killing the dancer didn't matter.

It's so fucking funny, there's way more than enough ambiguity to make the audience doubt what happened, there was DEFINITELY enough ambiguity to make Tony not just kill someone with his bare hands in a fit of rage, he just does it anyway because he's a dumbfuck.

I don't give a FUCK what David Chase thinks. The co-writers found it so ambiguous that they couldn't even agree on what happened and the actor was playing a not-guilty Ralph on screen.
ALL that matters is that whether he killed the horse or not is the option that makes the narrative more compelling and interesting, and the story is SIGNIFICANTLY better if he didn't kill the horse.
